Gladys or Aunt Gladys is a close friend of James James and served with him in the Spartan 1.0 program. Even though Janissary James refers to her as "Aunt" it is never confirmed that they're blood relatives and she could just be a close friend of Jan's father. In break_em.wav Gladys says she outranks James James.
When Jan finds out that she's tweaked and is a Spartan 1.1 she gets Gladys to take her to a Moon bar to test her newly discovered skills.[1] It's at that bar that she meets Thin Kinkle which leads to her getting kidnapped and James James being killed.
