DISCLAIMER: Halo 3 is, at this moment, NOT YET officially confirmed by developer Bungie Studios.  It is expected to appear as a title for the XBOX 360, possibly concurrently with the Halo Movie in development at Universal Studios. The game was announced on April 4th, 2005 on www.gamespot.com, but any information coming from sources not affiliated with Bungie Studios is pure speculation.
As of May 9, 2006, Halo 3 has been announced for release in 2007 on the Xbox 360.
Cortana's line in the trailer: "This is the way the world ends," references the first of the Cortana Letters. The line itself refers to T. S. Eliot's The Hollow Men, which ends:

Halo 2's Plot Developments
:This is the way the world ends
:Not with a bang but a whimper.

It is revealed in Halo 2 that all other Halo rings in the galaxy have been activated and put into standby mode, and that only a device known as The Ark has the capability to shut them all down. Sergeant Johnson, Miranda Keyes, 343 Guilty Spark and the Arbiter ponder this in the Halo control room. It is inferred that this device was sequestered away by the Forerunners, probably somewhere on Earth in remote pre-history. The Master Chief, meanwhile, returns to Earth in a spaceship he stowed away on when he left Cortana behind at her insistence. The ship that the Chief is on is a Forerunner Ship and it is occupied both by the last Prophet and its AI, which Cortana said she had great difficulty with as it fought her. Both the Master Chief and the Prophet are probably trying to head to the location of the Ark on Earth. The stage is set for a climactic final battle on Earth to find and gain control of the Ark which has either the power to shut down or resume the Halo Installation superweapon firing sequence all over the galaxy and also on High Charity to re-defeat the Flood.
Cortana's emphasis on "this" implies she is rejecting the poem, and that the world will end with a bang.

After the credits, we are treated to a small cinematic. The short movie reveals the omnipresent nature of the controlling intelligence behind the Flood - the massive creature that lives within its depths known as the Gravemind. He recites a verse:
It is also obvious that the MA5B Assault Rifle (looks some way like a Battle Rifle also) makes an appearance. Other vehicles such as the Phantom, Banshee, and the Covenant Assault Carrier appear in the trailer.

:Silence fills the empty grave now that I am gone,
The structure in the trailer is thought to be, but not officially confirmed as, The Ark. It was mentioned in Halo 2 as a place where all the Halo rings could be fired at once, and "This is the way the world ends" most likely refers to the Halo's activation. Though it has not been confirmed or denied by Bungie, it is seen in this small sketch from the DVD. Also, in Bungie's FAQ, they said that although they wouldn't tell what the structure is, Halo fans may recognize it. The structure looks somewhat similar to a structure seen in the Halo 2 multiplayer map Ascension or the circular structures on the exterior of Halo which is visible in the opening menu of Halo 1. Master Chief's gloves and armor have changed noticeably.
:But my mind is not at rest, for questions linger on.
:I will ask and you will answer.

Cortana appears above a console; its tentacle reaches out to her, but she raises her hand to halt its grasp - "Alright, shoot." is her response. Interestingly, as the Gravemind speaks in verse, the iambic pentameter of this phrase suggests that it is annoyed or worried.
"The details of the plot have yet to be revealed. Suffice to say, the Master Chief has returned to Earth. The Covenant has uncovered a massive and ancient artifact under the African soil and Cortana is still trapped in the clutches of the Gravemind." --Bungie.net

Speculation
Although some fans believe the trailer to be what Halo 3 will look like, the trailer was mainly to gain interest in the game.
The Rampancy of Cortana
Cortana's agreement to answer the Gravemind's questions leaves open the possibility that Cortana may have become - or may be becoming -  Rampant, and either giving some information to or forming some sort of alliance with the Flood which will join the final battle for control of the Ark.

It should also be noted that Cortana is not a biological lifeform but a computer program and would likely be unaffected should Halo actually fire. It is likely that Cortana is aware of this, so people should factor that into her reasoning.
"The trailer was built to have minimum impact on the development process, and while it required long hours and hard work from many, many Bungie staffers, it utilizes real-game assets, fiction and locations from parts of the "real" game."

Cortana going Rampant would also be in keeping with the tradition of other rampant AI's in Bungie games - ie: the Marathon series. It appears at least that Cortana has motivations that aren't entirely clear and may in fact be above and beyond her programming.
In the first Bungie weekly update after the trailer, the location is confirmed. The scene in the trailer is at the edge of New Mombassa. The mountain in the background is indeed Kilamanjaro. Also, the weapon the Chief is holding has been confirmed as an Assault Rifle, by Bungie, altough they state that it is not the same as the Assault Rifle in Halo 1. They have also said that it will be in the final game.

There are various references to Cortana's state of mind (rampancy) in the Fall of Reach, at the beggining of the section 'Two betrayals'. It is noted in this that she has spent a long time in a huge network - In keeping with Marathon's guide to rampancy.

Master Chief's Shields
*Bungie has confirmed via their Weekly Update at Bungie.net that multiplayer as well as singleplayer testing are currently in progress for the title, in varying stages of completion.
*The Bungie website also alluded to "a devastated and Covenant controlled Earth".
*Bungie revealed that the game would have a new "materials system" so advanced that "you could see light reflect off the ground and onto his armor", which will be able to "[show] the difference between his armor and the rubber undersuit". The site also includes that "It's so detailed that if you bring the camera close to the Master Chief's visor, you see everything in front of him reflected in it, right down to the ammo counter in his rifle" . In addition, there would be new advanced texturing to reflect permanent, persistent damage.
*In the trailer, Master Chief's Mark VI armor is battle-worn and plasma-scored. Developer Marcus R. Lehto's response: "The Chief is shown as warrior who has seen horrific battle – and it shows. The Earth is dry, barren, ravaged by the Covenant."
*Bungie has confirmed the following basic plot points from what the teaser trailer revealed:  Master Chief is back on a thoroughly conquered and Covenant controlled Earth.  Cortana is still a prisoner of the Gravemind.  Finally, the Covenant ships are hovering over an excavation site only a few miles from New Mombasa.
*Bungie has also stated that the rising structure seen in the trailer should be familiar to Halo fans.
*The Master Chief from Halo and Halo 2 is confirmed to be the Spartan in the Halo 3 trailer. Bungie has yet to release any information on the whereabouts of other Spartan soldiers mentioned in the Halo novels.
*Halo 3 and the Halo Movie could come out at the same time, however "The two will not affect each other in terms of release dates."
*Although Halo 3 was planned to end the Halo series, future Halo games haven't been entirely ruled out. On the Halo 3 FAQ from the official Bungie website it says, "Q: Is this the last ever Halo game? A: It is the conclusion to this story arc." Fans note that it is therefore possible that future games could be produced in the Halo universe, even if this particular story arc isn't continued.

In the previous two Halo games, the earliest actions that the player undertake in the guise of the Chief include recharging of shields. The Chief has not had an opportunity to recharge his shields since he left Cairo Station in Halo 2. With this in mind, it is likely that the Chief will start out with shields at less-than-maximum charge, and only regain the maximum charge for his shields once he returns to a human-controlled space and gets to a recharger, or otherwise hijacks a Covenant-made recharger for his use.

Another fan argues:

Actually, that is completely wrong. That device is used to "test" the chief's shields, to see if they are in working condition. In the first game, he came out of cryo sleep, and so they needed to test if the cryo had affected his shields at all. In Halo 2, Master Chief gets brand new armor, and since it is brand new, it obviously needs tested.

First Level
It has been speculated by many people that the first level may acctually remble the New Mombasa fight that was origanly planed for Halo 2, it is also belived that it will feature a mini lvl of Master Chief taking control of the Forerunner artifact and having to land it some were near the battle going on at New Mombasa. [or possibaly link up with Frederic and learn the wearabouts of the other Spartans]

More Spartans
The possibility of the other Spartans return is also highly possible. There is a chance that Kelly will come back stronger due to her time with Dr. Hasley and the Forerunner artifact, as well as the fact that Linda, Will, and Frederic will fight by John early on or later in the game.

Cortana's emphasis on "this" implies she is rejecting the poem, and that the world will end with a bang.

Although some fans believe the trailer to be what Halo 3 will look like, the trailer was mainly to gain interest in the game.
